摘要： Lung adenocarcinoma (LUAD) remains a leading cause of cancer-related mortality, largely due to its aggressive behavior and early metastatic potential. In this study, FOS-like 2 (FOSL2) was identified as a transcriptional regulator that upregulates L1 cell adhesion molecule (L1CAM) in LUAD. L1CAM was found to be upregulated in LUAD cell lines compared to non-malignant lung epithelial cells. Functional assays demonstrated that silencing L1CAM reduced cell proliferation, migration, and invasion, increased apoptosis, and inhibited activation of the PI3K/AKT/mTOR pathway. Suppression of FOSL2 produced similar effects in vitro and also limited tumor growth in vivo. Treatment with the AP-1 inhibitor T5224 reduced proliferation of A549 cells. Moreover, knockdown of L1CAM attenuated the tumor-promoting effects of FOSL2 overexpression, indicating that L1CAM contributes to the oncogenic activity of FOSL2, possibly by mediating activation of the PI3K/AKT/mTOR signaling pathway.

摘要： This study presents the first genome-wide identification and comprehensive analysis of laccase (LAC) and cinnamyl alcohol dehydrogenase (CAD) gene families in Populus simonii, revealing 39 PsiLAC and 18 PsiCAD members. Phylogenetic and structural analyses classified these genes into distinct subfamilies, highlighting conserved motifs and evolutionary divergence. Tissue-specific expression patterns demonstrated high activity in roots, leaves, and apex, while abiotic stress responses implicated their roles in drought and salt tolerance. Since the knockout of COMT leads to the accumulation of the C-lignin precursor (caffeyl alcohol), we chose the COMT-KO line as the background. Additionally, research results on Cleome hassleriana showed that CAD5 has a higher affinity for caffealdehyde and can specifically catalyze the reduction of caffeoyl-CoA (caffeoyl coenzyme A) to caffeyl alcohol. This allows LAC8 to promote the polymerization of caffeyl alcohol to form C-lignin. A groundbreaking achievement was the construction of a PsiLAC8-PsiCAD5 tandem gene module, which was transformed into COMT-KO poplar lines. Transgenic plants exhibited elevated lignin content and altered phenotypes, providing functional evidence for their involvement in lignin polymerization. This work not only advances understanding of lignin biosynthesis in woody plants but also establishes a foundation for engineering C-lignin production in Populus, offering novel strategies for improving biomass utilization and stress resilience. The integration of bioinformatics, expression profiling, and transgenic validation underscores the dual functional significance of PsiLAC8-PsiCAD5 in lignin synthesis and stress adaptation.

摘要： Cracks pose a major threat to the safety and durability of hydraulic concrete structures. However, existing crack segmentation methods often suffer from crack disconnection, blurred boundaries, over-segmentation in complex environments, and high annotation costs. To address these issues, this paper proposes a weakly supervised crack segmentation method—Dual-Stage Refined Class Activation Map with Ivy-optimized Dense Conditional Random Field (DSRCAM-ICRF)—which achieves high precision using only image-level labels. The framework consists of two stages. In Stage I RCAM, Convolutional Neural Network (CNN) and Transformer branches extract local and global features. In Stage II RCAM, attention weights from different layers are scaled and fused to account for depth-wise variation, enabling precise integration of hierarchical features and enhancing fine crack localization. The Ivy-optimized DenseCRF module further refines crack boundaries and mitigates over-segmentation. Experiments on UAV-captured datasets demonstrate superior performance, with a Mean Intersection over Union (MIoU) of 0.823, precision of 0.908, and F1-score of 0.901. This method offers a robust and scalable solution for automated crack detection in structural health monitoring.

摘要： Conventional bone tumour treatment often results in the disruption or collapse of the structural integrity of bone following tumour resection, while the accompanying application of chemotherapy frequently results in wide systemic damage due to toxicity. Such shortcomings in current bone tumour treatment options highlight the need for the development of multifunctional biomaterials that serve as systems for targeted drug delivery and biodegradable support, stimulating new bone formation. In this work, chitosan-calcium phosphate composite scaffolds were used as carriers for local delivery of antitumour drugs and therapeutic agents, while providing bioactive properties. The bioactivity of composite scaffolds was confirmed by an apatite-rich layer formation in an acellular in vitro test, and by osteogenic differentiation of human mesenchymal stem cells cultured for 21 days. The encapsulation of an antitumour drug, doxorubicin, was achieved through chemical crosslinking by genipin, while its release was characterised in a complete cell culture medium. The rapid release of doxorubicin affected the viability of human dermal fibroblasts (HDFa) and osteosarcoma cells (MG-63) when in contact with drug-loaded scaffolds. Following osteogenic properties, the modification of the composite scaffold with boric acid was also explored. The rapid release of boric acid from the composites was reported to be within the first three days of incubation, which stimulated a slight increase in the expression of vascular endothelial growth factor in HDFa cells. A biotolerability study was performed by chorioallantoic membrane (CAM) assay, indicating tissue infiltration into composite scaffolds with a lower concentration of boric acid. The rapid delivery of doxorubicin or boric acid from biodegradable and osteoconductive composite scaffolds shows the potential to address the clinical challenge of local tumour recurrence and poor bone regeneration following resection.

摘要： In recent years, powered airships have been considered promising alternatives for high-altitude, long-endurance missions. To address the issues of high computational costs, insufficient consideration of engineering constraints, and disconnect from manufacturing validation in existing global optimisation methods, a local optimal multidisciplinary design optimisation (MDO) framework based on genetic algorithms (GAs) is proposed in this study. In this framework, an industry-friendly lightweight parametric model (seven-segment Bézier) is innovatively used for parametric modelling of the airship fuselage, which enables precise control over critical geometric features. By establishing a multiobjective fitness function with adjustable weights and incorporating nonlinear constraints, the multi-objective genetic algorithm (MOGA) is used to efficiently search for locally Pareto optimal solutions within the feasible engineering domain. The optimised results are reconstructed into a three-dimensional model in SolidWorks and validated rigorously through a high-fidelity virtual flight test platform. Compared with the baseline design, the optimised scheme achieves a significant increase in volume of 29.8 % and 9.5 %, a reduction in the drag coefficient of 7.2 % and 4.3 %, and an improvement in energy efficiency of 26.4 % and 4.8 % while maintaining the structural stress threshold. Sensitivity analysis and Monte Carlo simulations further confirm the robustness of the solution to manufacturing tolerances. The integrated framework established in this study, which combines parametric modelling with GA local optimisation and CAD/CFD validation, provides a computationally efficient, reliable, and manufacturable solution for the engineering design of stratospheric airships, thereby significantly advancing the transition from theoretical optimisation to practical application.
