摘要： In this work, a novel copper (I) catalyst was heterogeneously immobilized onto acrylic fiber functionalized with a bidentate amidine bearing chelating ligands as a robust support. The amidine-modified fiber was prepared via a two-step process involving covalent attachment of ethylenediamine to acrylic fiber followed by condensation with acetamide. Subsequent coordination of copper (I) iodide onto the modified fiber resulted in the catalytically active complex PANF_Amd@Cu (I). The catalyst was characterized by FTIR, diffuse reflectance UV–vis spectroscopy, XRD, FE-SEM, EDX, TG/DTGA, and elemental analysis. Its catalytic performance was tested in one-pot, three-component click cycloaddition reactions of alkyl halides, sodium azide, and terminal alkynes in aqueous media. The heterogeneous catalytic system demonstrated high regioselectivity toward 1,4-disubstituted 1,2,3-triazoles with excellent yields in relatively short reaction times. The amidine linkage in the fiber matrix effectively stabilized the active Cu (I) species, significantly enhancing catalytic efficiency. Moreover, the catalyst could be conveniently recovered by simple mechanical means (e.g., tweezers) and reused for multiple cycles with minimal loss in performance and negligible copper leaching, demonstrating sustainability and eco-friendliness. This work highlights the potential of functionalized acrylic fibers as sustainable, cost-effective, and recyclable supports for heterogeneous Cu (I) catalysts in environmentally benign organic synthesis.

摘要： Imposing an electric field could intensify reactions under mild conditions. This study proposed to enhance alkane cracking for light olefins production, one typical process requiring harsh conditions, by electric field in the catalysis by Zr-doped CeO₂ (Ce_xZr_1−xO₂). It was found that Zr incorporation significantly enhanced oxygen storage/release capacity of CeO₂. Spectroscopic analysis revealed that electric field induced lattice distortion in the (2 0 0) plane, and the resulting elongated Ce-O bonds facilitated lattice oxygen release. Under the reaction conditions of electric power of 10 W with external heating at 400 °C, n-hexane conversion reached 59.3% and light olefins selectivity was 67.4% over Ce_0.50Zr_0.50O₂. These results markedly outperformed the values obtained in the conventional thermal mode (n-hexane conversion of 0.5% and light olefins selectivity of 13.1% at 450 °C). And it was found that the n-hexane conversion increased linearly with the increase of migrated lattice oxygen content in the catalyst. DFT calculations show that the electric field optimized the d-p orbital hybridization between Ce_0.50Zr_0.50O₂ and n-hexane, enhanced the adsorption of reactant on the catalyst, and accelerated the charge transfer between the catalyst and reactant. In situ synchrotron radiation vacuum ultraviolet photoionization mass spectrometry (SVUV-PIMS) demonstrated that the electric field could accelerate n-hexane dehydrogenation to form more hexene and trigger the formation of free radicals (e.g., C₂H₅·) under 400 °C. All the above led to efficient n-hexane cracking to light olefins, especially ethylene, under an electric field at mild conditions. This work provides an alternative strategy for the low-carbon transformation of the petrochemical industry.

摘要： 1-Phenylethylamine (PEA) is an important chiral intermediate in pharmaceutical synthesis and fine chemical production, and its two enantiomers often show markedly different biological activities. Therefore, efficient enantioselective recognition of PEA is of considerable practical importance. In this work, a chiral inducer-analyte congruent strategy was adopted to build enantioselective sensing materials while avoiding the introduction of additional non-chiral structural variables. Using (R)-1-PEA and (S)-1-PEA as non-embedded chiral inducers, a pair of homologous mirror-image homochiral TpPa-1 covalent organic frameworks (COFs), denoted as (Δ)-TpPa-1 and (Λ)-TpPa-1, was successfully prepared. Both COFs exhibited distinct mirror-image circular dichroism (CD) signals together with well-defined helical morphologies. After being integrated onto quartz crystal microbalance (QCM) chips, the resulting dual-channel sensing platform showed configuration-matched responses toward PEA enantiomers, with maximum enantioselectivity factors of 1.65 and 2.59 for (Δ)-TpPa-1 and (Λ)-TpPa-1, respectively, and a linear range of 10–60 μM. The high selectivity can be ascribed to the chiral memory effect established during framework formation and to the conformational complementarity between the chiral COF channels and the target enantiomers. This study provides a straightforward strategy for designing homochiral COF-based sensors for chiral amine recognition.

摘要： Dysfunctional tumor vasculature limits immune checkpoint blockade efficacy. We have recently established that endothelial SPEN deficiency promotes tumor vessel normalization by suppressing RNA polymerase I (RNAPI)-mediated rDNA transcription via inducing disrupted ribosome biogenesis and nucleolar stress, but whether this pathway could be practically employed in immunotherapy has been unclear. In the current study, analysis of publicly available single‑cell RNA‑seq datasets revealed that endothelial SPEN expression was downregulated in clinical responders, including colorectal cancer patients achieving pathological complete response (pCR) after PD‑1 blockade and lung adenocarcinoma patients showing major pathological response (MPR) after neoadjuvant PD‑1 blockade plus chemotherapy. Using endothelial-specific SPEN knockout mice (eSPEN^−/−), we demonstrate that SPEN loss in established tumors remodels the immune microenvironment by enhancing the infiltration and perivascular accumulation of both CD8⁺ and CD4⁺ T-cells, and in addition, boosting the effector function of CD8⁺ T-cells. A combined therapy of anti-PD-1 with CX-5461, a small-molecule inhibitor of RNAPI yielded better tumor inhibition than monotherapy with anti-PD-1 alone. This was attributable to increased T-cell infiltration and a marked expansion of polyfunctional CD8⁺ T-cells capable of producing both IFN-γ and TNF-α. In summary, our study identifies endothelial RNAPI, which is regulated by SPEN, as a target of normalizing tumor-immune microenvironment and that the RNAPI inhibitor, which is under clinical trial, is potentially useful to improve PD-1 blocker in cancer therapy.

摘要： The reconstruction of the runaway electron (RE) energy distribution is critical for understanding RE dynamics and mitigation in tokamak plasmas. In this work, the energy distribution of runaway electrons in a low-density Ohmic discharge of the Thailand Tokamak-1 (TT-1) was determined by unfolding hard X-ray (HXR) spectra measured using a LaBr${}_3$(Ce) scintillation detector. To address the ill-posed inverse problem caused by the complex detector response, a comprehensive response function matrix was constructed using FLUKA Monte Carlo simulations, incorporating the detailed TT-1 geometry and the detector. Two statistical unfolding algorithms, the iterative Bayesian method and Maximum Likelihood Estimation (MLE), were applied to experimental data from a representative low-density discharge. The reconstructed spectra from both methods showed excellent agreement, with a relative difference on the order of $10^{-4}$ %. The results suggest the energy distribution of runaway electrons centered around 6.5 MeV. This study establishes a robust methodology for RE energy diagnostics in TT-1, providing a necessary framework for future transport and loss mechanism studies.

摘要： Macrophages are primary regulators of the innate immune system, integrating pattern recognition receptor (PRR) signaling and cytokine production to shape early inflammatory processes. However, the molecular mechanisms governing macrophage-driven responses in chickens remain poorly understood, despite their importance in controlling major infectious diseases. Among these pathways, interleukin (IL)-1β functions as a central mediator of inflammatory signaling, yet its integration with Toll-like receptor (TLR) pathways in chicken macrophages remains unclear. Here, lipoteichoic acid (LTA)-Toll-like receptor (TLR)2, poly(I:C)-TLR3, and lipopolysaccharide (LPS)-TLR4 pathways were investigated in the chicken macrophage-like cell line HD11 and an IL1β-deficient HD11 mutant, with PBMC-derived macrophages included for comparison. HD11 cells exhibited minimal responsiveness to LTA–TLR2 stimulation, whereas LPS–TLR4 stimulation elicited a limited response primarily characterized by strong induction of IL-1β and IL-8 but minimal or undetectable IL-6, tumor necrosis factor (TNF)-α, IL-18, and interferon (IFN)-β. In contrast, poly(I:C)–TLR3 stimulation induced IL-1β, IL-18, and IFN-β with limited induction of other cytokines. A genetically engineered CRISPR/Cas9 IL1B knockout (KO) HD11 model has been established to evaluate molecular interactions of the inflammatory response, which appears to be modulated by IL-1β in a TLR ligand-specific context. The present study highlights distinct pattern recognition profiles on chicken macrophages that may offer a molecular basis for IL-1β-targeted signaling networks and immunomodulation in chickens.

摘要： Acute hyperglycemia in ischemic stroke occurs in almost half of cases,inducing a worsening of the underlying pathophysiology.Hence,the management of acute hyperglycemia during the first hours following ischemic stroke requires well-defined strategies.Furthermore,the effect of hyperglycemia on stroke remains an unresolved paradoxical process whose possible causes are still unknown.The ischemic process involves the activation of multiple signaling pathways related to cell death and survival,which can be acutely altered by hyperglycemia,causing subsequent worsening of the ischemic process.In search of potential biological targets to combat this medical burden,the role of hypoxia-inducible factor appears as a novel neuroprotective agent that needs further investigation.Furthermore,hypoxia-inducible factor protein targets multiple pathways changing depending on factors such as the severity of hypoxia,the time elapsed after reperfusion and the specific cell type.In fact,the poor prognosis of ischemic stroke associated with acute hyperglycemia has been linked to alterations in hypoxia-inducible factor-1αprotein function.Together,these findings support this transcription factor as a new therapeutic target to prevent the negative effect of hyperglycemia,opening new avenues for treatment in stroke.
