摘要： Liver cirrhosis is a chronic liver disease characterized by persistent hepatic injury and progressive fibrosis, accompanied by increased apoptosis and inflammatory responses. Although the polysaccharide GM-1, derived from Rhizoma Amorphophalli, has demonstrated immunomodulatory properties, its role in liver cirrhosis remains poorly understood. In this study, GM-1 was extracted and purified from Rhizoma Amorphophalli, and its structural features were systematically analyzed. Using a comprehensive approach integrating network pharmacology, transcriptomic analysis, a TNF-α-induced AML12 hepatocyte model, and a CCl₄-induced SD rat model, the hepatoprotective effects and potential mechanisms of GM-1 were explored. The results revealed that GM-1 has a molecular weight of 57,520.48 Da and is predominantly composed of mannose and glucuronic acid, with a main chain structure consisting of →4)-β-D-Manp-(1→, →4)-β-D-Glcp-(1→. GM-1 may alleviate liver cirrhosis, potentially through the modulation of key signaling pathways, including IL-17 and NF-κB, thereby suppressing inflammation and apoptosis and reducing hepatocellular damage. This study provides a systematic understanding of the protective effects and potential molecular mechanisms of GM-1 in liver cirrhosis, offering valuable experimental evidence for further evaluation of its therapeutic potential.

摘要： Effective cancer therapy hinges on the ability to selectively target malignant cells while sparing healthy tissues. For virotherapy this goal can be achieved by genetic engineering the surface glycoproteins of viruses. This approach, though, often leads to loss of functionality of the engineered proteins, in terms of membrane fusion. Here, we modified the E2 protein of Sindbis virus, preserving the ability of transduction by lentivectors pseudotyped by the Sindbis Env proteins. The modifications introduced allowed to alter the efficiency of transduction of glioblastoma cells expressing integrin α5β1, a driver of resistance to treatments and frequently associated to recurrences. Importantly, the same modifications of E2 also conferred, to a replication-competent Sindbis strain, the ability to kill preferentially α5β1⁺ over α5β1⁻ glioblastoma cells in culture. Finally, the same strain reduced the expansion of α5β1⁺ glioblastoma cancers in the brain of immunodeficient mice when injected in the tail vein.

摘要： 探究“1+X”证书制度背景下高职机电一体化专业课程模块化教学模式,旨在优化教学的个性化设计与灵活配置,提升学生跨学科知识整合能力。以数控加工编程课程为例,分析当前专业课程教育现状,发现存在教学内容单一、教学方式陈旧等问题,在此基础上提出课程模块化教学的制定思路,明确核心课程应围绕“1+X”证书制度下的教学目标,契合实际工作岗位要求,遵循因材施教原则,贴合学生发展需求,从而更好地引领职业教育的课堂革命。

摘要： 目的探讨血清肾损伤分子1(KIM-1)对急性心肌梗死(AMI)患者发病后72 h内急性肾损伤(AKI)早期危险分层的潜在价值。方法连续纳入2025年11月至2026年1月于首都医科大学附属北京安贞医院住院的30例AMI患者开展单中心前瞻性初步研究。依据是否发生AKI分为AKI组(10例)和非AKI组(20例)。于入院即刻(T_(0))、入院后6~12 h(T_(1))、入院后24 h(T_(2))、入院后48 h(T_(3))、入院后72 h(T_(4))采集血清,检测KIM-1与血肌酐水平,采用受试者工作特征曲线分析其预测72 h内AKI的价值。本研究为探索性研究,样本量有限,结果仅用于提出假设并报告置信区间。结果AKI组T_(0)时点KIM-1水平显著高于非AKI组[0.150(0.095,0.305)μg/L比0.080(0.058,0.132)μg/L](P=0.019);T_(0)时点KIM-1预测72 h内AKI的曲线下面积(AUC)为0.77(95%置信区间:0.52~0.94),优于同期血肌酐(AUC=0.66,95%置信区间:0.45~0.88)。以0.100μg/L为临界值,KIM-1预测AKI的敏感度为100%(95%置信区间:69%~100%)、特异度为60%(95%置信区间:36%~81%)。在2组从症状发作到入院的中位时间一致情况下,患者KIM-1于T_(0)时点即表现组间差异,血肌酐直至T_(2)时点才出现差异,二者时间差约24 h。结论入院时血清KIM-1对AMI后AKI发生具有早期危险分层的潜在价值,检出时间早于血肌酐,但其探索性结果需大样本、多中心研究外部验证及临床实用性评估后,方可转化应用。

摘要： Pulmonary fibrosis can result from autoimmune, inflammatory, or idiopathic conditions, including systemic sclerosis-associated interstitial lung disease (SSc-ILD). Previous studies have implicated myofibroblasts and SPP1^hi macrophages as drivers of fibrosis in lungs, as well as other tissues. Single-cell RNA-sequencing has delineated fibroblast and macrophage transcriptomes but provides limited insight into the transcriptional control of profibrotic gene programmes.
