摘要： The increased utilization of glucagon-like peptide-1 receptor agonist (GLP-1 RA) medications to manage diabetes mellitus and weight loss has raised questions about their impact on total shoulder arthroplasty (TSA) outcomes. Studies of GLP-1 RA effects on total hip and knee arthroplasty have reported a decrease in 90-day readmission rates and perioperative joint infections, suggesting that GLP-1 RA patients had better outcomes. The data are less clear for TSA patients as conflicting studies have shown no effect on outcomes while others suggest a protective effect from GLP-1 RA use. The purpose of this study is to determine the effects of GLP-1 RA usage in diabetic patients undergoing TSA on clinical outcomes.

摘要： Postharvest tomato fruit are susceptible to gray mold caused by Botrytis cinerea (B. cinerea) infection, which impairs the quality and marketable yield of the fruit commodity and thus undermines the economic sustainability of the tomato industry. This study was designed to explore the control efficacy and underlying mechanisms of Trichoderma harzianum (T. harzianum) strain NDT-1 against postharvest gray mold in tomato fruit. Physiological and biochemical assays indicated that pretreatment with NDT-1 enhanced defense enzyme activity, mitigated oxidative stress, and regulated the contents of defense related hormones. NDT-I triggered intricate calcium (Ca^2 +) signaling pathways that regulate calcium dependent protein kinase (CDPK) and respiratory burst oxidase homolog (Rboh) gene expression. These pathways constrained excessive reactive oxygen species (ROS) accumulation and alleviated oxidative damage. Concurrently, immune signaling involving mitogen-activated protein kinase 3 (MPK3), pathogen-induced transcription factors (Pti5/6), and WRKY transcription factor 22 (WRKY22) induced the expression of defense genes, specifically pathogenesis-related protein 1 (PR-1). Additionally, whole-transcriptome analysis revealed that non-coding RNAs (ncRNAs) play a part in post-transcriptional regulation, including MSTRG.2359.2, MSTRG.13844.1, MSTRG.16844.3, and SL4.0ch11_ 53096625_53108375. Collectively, these findings suggest that NDT-1 is a promising biocontrol agent for postharvest gray mold in tomatoes.

摘要： Lung ischemia-reperfusion injury (LIRI) is a severe complication of lung transplantation and acute respiratory distress syndrome, leading to significant pulmonary dysfunction. Aloperine, a natural alkaloid derived from Sophora alopecuroides, has demonstrated promising pharmacological effects on various ischemia-reperfusion injuries. However, its role in LIRI remains underexplored. Our experimental results revealed that aloperine significantly alleviated bilateral lung tissue damage induced by ipsilateral ischemia-reperfusion, with a mechanism closely linked to the regulation of inflammatory responses and apoptosis-related proteins. Further studies demonstrated that aloperine specifically activated the STAT-1 signaling pathway, as evidenced by markedly increased STAT-1 phosphorylation levels in lung tissue damage in the ipsilateral (ischemic) and contralateral (non-ischemic) lung, thereby inhibiting ferroptosis. Notably, the STAT-1 inhibitor AG490 completely reversed the lung-protective effects of aloperine, confirming the protective effects of STAT-1. These findings provide critical experimental evidence for the potential clinical application of aloperine in LIRI intervention.

摘要： Myocardial infarction (MI) and stroke are examples of ischaemic diseases that continue to pose serious health risks since few specific treatments address the underlying molecular causes of these conditions, which include pathological thrombosis and mitochondrial dysfunction. In order to close this gap, we used an integrated in-silico method to find strong inhibitors for Protease-Activated Receptor-1 (PAR-1) and Cyclophilin D (CypD) from the structurally diverse CMNPD marine natural product library. To account for protein flexibility, our method combines High Throughput Virtual Screening (HTVS), Standard Precision (SP), and Extra Precision (XP) docking with meticulous Induced Fit Docking (IFD). After undergoing 500 ns Molecular Dynamics (MD) simulations, the best candidate compounds showed remarkable structural stability with RMSD values between 0.6 and 1.7 Å. Strong binding affinities were found by thermal MM-GBSA calculations, with CMNPD29955 showing a ΔG of −63.146 kcal/mol towards CypD and CMNPD3572 and CMNPD22037 showing ΔG values of −96.24 and −98.092 kcal/mol against PAR-1, respectively. Lead molecules of each target were found to have important hydrophobic and hydrogen bonding interactions with crucial active site residues in MD modelling. The dynamic stability and energetic favorability of these chemicals were further confirmed by complementary techniques, including WaterMap, Principal Component Analysis (PCA), and Free Energy Landscape (FEL). In addition to CMNPD3572 and CMNPD22037 as potential PAR-1 antagonists, this work emphasises CMNPD29955 as a new lead inhibitor that targets mitochondrial dysfunction via CypD. To transform computational insights into effective and complementary therapeutic alternatives, lead optimisation, experimental validation, and preclinical assessment of these special marine-derived molecules will be crucial in the future.

摘要： Magnetic covalent organic framework nanocomposite denoted as Fe_(3)O_(4)@TPBD-BTA with core-shell structure was fabricated via a simple template-mediated precipitation polymerization method at mild *** polyimine network shell was created through the polymerization of N,N,N’,N’-tetrakis(p-aminophenyl)-pphenylenediamine(TPBD)and biphenyl-3,3’,5,5’-tetracarbaldehyde(BTA)in tetrahydrofuran by the Schiffbase *** with large good solution dispersibility and high stability,the obtained Fe_(3)O_(4)@TPBD-BTA exhibited high adsorption capacities and fast adsorption for aflatoxins(AFs).With the Fe_(3)O_(4)@TPBD-BTA as sorbent,a magnetic solid-phase extraction high performance liquid chromatography-tandem mass spectrometry method was established for simultaneous adsorption and detection of two AFs in complex *** proposed method displayed favorable linearity,low limits of detection(0.0065-0.0082μg/kg).The developed method has been applied for real sample analysis,with recoveries of 81.3%-87.4%.These results showed that Fe_(3)O_(4)@TPBD-BTA has a good application potential for the adsorption of AFs in milk and dairy products.

摘要： The weak ozone spectrum is recorded by cavity ring down spectroscopy in the region of the Wulf band near 1 µm. The line profile of six regular sequences of mostly resolved lines belonging to the singlet-triplet ³A₂(010)-X¹A₁(000) band are studied between 9760 and 9915 cm^-1. The upper states of the 47 targeted transitions are affected by predissociation processes due to the mixing of the ³A₂ upper triplet electronic state with the near lying states of the X¹A₁ ground state, by spin-orbit coupling. A series of four recordings for ozone pressures ranging between 10 and 40 Torr allows for (i) disentangling the pressure broadening (proportional to pressure) and the (dominant) predissociation broadening (independent on pressure) (ii) determining accurate values of the absolute line intensities, on the order of 10^–24 cm/molecule for the strongest transitions. The retrieved predissociation widths range between 0.03 and 0.37 cm^-1 corresponding to lifetimes of 87 and 7 ps, respectively. The magnitude of these values and their strong dependence on rotational quantum numbers are discussed.
